The kinetic behavior of mushroom tyrosinase was studied during the oxidation of l-tyrosine and l-DOPA. Under the conditions employed in the present study, the progress curves for the enzyme at different concentrations of l-tyrosine and l-DOPA
have been determined. When the diphenolase activity of mushroom tyrosinase was assayed by using l-DOPA as substrate, the reaction immediatelyreached a steady-state rate (Fig. 1, curve 1). It was a straight line which passed through the original
point of the coordinate, and the slope of the straight line was the initial rate (v0). The relationship of the initial rate and the concentration of substrate (l-DOPA) followed Michaelis–Menten kinetics. Kinetic parameters for the diphenolase activity of
mushroom tyrosinase are shown in Fig. 4 (line 0) as Lineweaver–Burk plot. The apparent Michaelis constant (KDm) is 0.68±0.02mM and the apparent maximum velocity (VDm) is 151.9±2.5M/min. When the monophenolase activity of mushroom tyrosinase was
assayed by using l-tyrosine as substrate, a remarkable lag period, characteristic of monophenolase activity, was observed simultaneously with the appearance of the first stable product, dopachrome (Fig. 1, curve4). The system reached a constant rate after the

the reaction system reached the steady state, the curve of product increased straightly with increasing the reaction time. And the slope of the line denoted the steady-state rate. The relationship of the steady-state rate with the concentration of substrate
(l-tyrosine) followed Michaelis–Menten kinetics too. Kinetic parameters for the monophenolase activity of mushroom tyrosinase are exhibited in Fig. 5 (line0) as Lineweaver–Burk plot. The apparent Michaelis constant (KMm) is 0.29 ± 0.03mM and the apparent
maximum velocity (VMm ) is 36.5 ± 1.5M/min. |
